Thanx, my friends! After hours and hours I've found the error by myself:
Both kinds of climbing regions do work. What happend was, that there was a swim region that influenced the climbing region, even if it was not touching it.
After deleting the swim region the ladder worked again finally. Now I just have to create the swim region again and form it a bit different.
